چکیده

The intraoellular concentration of thymidine triphosphate has been measured in thymine requiring mutants of Escherichia co&i K12 and E. coli 15, and compared with that in non-mutant strains using thymine, thymidine and a mixture of thymine and deoxyguenosine, in the growth medium. The data show (1) that the thymidine triphosphate concentration rises as the thymine concentration in the growth medium is increased; (2) that the triphosphate concentration in both mutant strains is always less than that in the corresponding non-mutant strains; and (3) that thymidine inhibits its own conversion to thymidine triphosphate and also de nova synthesis of the triphosphate in non-mutant strains. It is known that in thymineless mutants the replication time of the chromosome increases ~CS the concentration of thymine in the medium decreases. The data presented in this paper suggest that the replication velocity is a function of, but not determined directly by, the intracellular thymidine triphosphete concentration.

Thymineless death in recombination deficient mutants of Escherichia coli K12.

When grown in thymine-free minimal synthetic medium, thyA recA mutants of Escherichia coli K12 were comparatively more resistant and thyA recBC mutants more sensitive to thymineless death (TLD) than their respective parent strains. No excessive numbers of single-strand breaks were observed in the DNA of the recBC mutant strain starved for thymine, hence the hypersensitivity for TLD in this stra...

Inhibition of thymidine transport in dnaA mutants of Escherichia coli.

DnaA protein is the initiator of chromosomal DNA replication in Escherichia coli. We report here our evidence that thymidine transport across cytoplasmic membranes in temperature-sensitive dnaA mutants is greatly decreased at a permissive temperature for growth of the mutants. Complementation analysis with a plasmid containing the wild type dnaA gene and P1 phage-mediated transduction confirmed...

Essential role for the Legionella pneumophila rep helicase homologue in intracellular infection of mammalian cells.

We have previously isolated 32 mutants of Legionella pneumophila that are defective in the infection of mammalian cells but not protozoa. The mutated loci have been designated macrophage-specific infectivity (mil) loci. In this study we characterized the mil mutant GK11. This mutant was incapable of growth within U937 macrophage-like cells and WI-26 alveolar epithelial cells. This defect in int...

Exonuclease VII of Escherichia coli.

A new exonuclease of Escherichia coli K12, exonuclease VII, has been purified 1700-fold and characterized. The enzyme is specific for single-stranded DNA and can initiated hydrolysis at both 5' and 3' termini. It is also capable of thymine-dimer excision in vitro. The limit products of the reaction are oligonucleotides, predominantly in the range of tetramers to dodecamers. DNA is hydrolyzed b...

Physiological and enzymatic properties of a thymidine-requiring Pediococcus cerevisiae mutant.

We describe the isolation and characterization of a Pediococcus cerevisiae thymidine-requiring mutant and its thymidine-independent revertant. The mutant strain lacked thymidylate synthetase activity and had an absolute requirement for low concentrations (2 micrograms/ml) of thymidine in addition to a requirement for N-5-formyl tetrahydrofolic acid (folinate). Even at high concentrations (up to...
